乳腺肿瘤端粒酶活性检测的临床意义 被引量:8

Clinical signiricance of measurement of telomerase activlty in human breast neoplasmas

摘要 目的探讨端粒酶在良、恶性乳腺肿瘤中表达的差异以及它与恶性乳腺肿瘤的病理分型、临床分期之间的关系。方法采用TRAP(telomericrepeatedamplificationprotocol)-银染法检测保存于液氮中的新鲜组织标本中的端粒酶活性。结果良、恶性乳腺肿瘤的端粒酶阳性率差异有显著性(P<0.005),但它们的瘤旁组织中的端粒酶活性皆为阴性。恶性乳腺肿瘤端粒酶的阳性率随病理分型的恶化和TNM分期的进展而逐渐增高。恶性乳腺肿瘤中肿瘤长径≥20mm及淋巴结转移阳性组的端粒酶阳性率显著高于<20mm及淋巴结转移阴性组(P<0.001)。结论术前检测乳腺肿块中端粒酶活性有利于良、恶性乳腺肿瘤的早期鉴别诊断和了解恶性乳腺肿瘤的发展状况,亦有利于乳腺肿瘤的早期治疗。 Objecteve To investigate the difference of telomerase expression in human benignbreast neoplasmas and malignant breast cancer and its relationship to the pathological types, clinicalTNM stages, growth and lymphatic metastasis of malignant breast cancer. Method The telomeraseactivity in fresh tissue preserved in liquid nitrogen was detected by using telomeric repeated amplification protocol silver staining assay. Results The positive rate of telomerase activity showed a significantdifference between malignant breast cancer and benign breast neoplasmas (P <0. 005). No telomeraseexpression was found in the adjacent noncarcerous tissues. Te1omerase activity was increased graduallywith the deterioration of pathological types and the development of TNM stages. The telomerase activity in malignant breast cancer with the long diameter of the tumor ≥20 mm and lymphatic metastasiswas higher than that in the group of the tumor diameter <2O mm and without lymphatic metastasis (P<0. 001). Conclusion The detection of the telomerase activity in breast neoplasmas before operationmight be helpful to early diagnose benifn breast neoplasmas and malignant breast cancer, to understandthe development of malignant breast cancer and to early treat the breast neoplasmas.
