一种分离自皱边石杉茎尖共生藻类的鉴定及培养

摘要 湖南产皱边石杉茎尖经过表面消毒和内生菌灭菌后,接种于愈伤组织诱导培养基中,结果分离培养出一种与其共生的藻类,经超显微结构鉴定为椭圆小球藻。扫描电镜结果显示,椭圆小球藻聚积成团状;透射电镜结果显示,椭圆小球藻细胞长7~12μm,宽4.5~8.0μm,椭圆小球藻的胞外多糖层明显分为两层,无性生殖时,原生质体直接分裂成4个似亲孢子,从母体中破壁而出。细胞内含有大量的淀粉核,色素体呈片状,占细胞较大部分;藻体内含叶绿素a 0.134 mg/g,叶绿素b 0.047 mg/g,类胡萝卜素0.057 mg/g,叶绿素a/b为2.855。 After the process of surface and endophytic bacteria sterilization,the were stem tips of Huperzia crispata were on the callus induction medium,separated and cultivated a kind of symbiotic algae which was called Chlorella ellipsoidea after identified by the microstructure.Scanning electron microscope show that the C.ellipsoide accumulated into a ball.Transmission electron microscopy show that the length of the cell was 7~12 μm,the width of the cell was 4.5~8.0 μm.The extracellular polysaccharide of C.ellipsoide had two layers,during its asexual reproduction,the protoplasts split into four autospores directly.C.ellipsoide cell contained a large number of pyrenoids,its pigment body was platelike.The chlorella contained chlorophll-a 0.134 mg/g,chlorophll-b 0.047 mg/g,carotenoid 0.057 mg/g,and the ratio of chlorophll-a to chlorophll-b was 2.855.
