摘要
目的观察舒冠滴丸对高脂血症大鼠缺血再灌注损伤心肌细胞间黏附分子-1(ICAM-1)表达的影响及其作用机制探讨。方法建立大鼠高脂血症模型,随机分为对照组、假手术组、模型组、辛伐他汀组及舒冠滴丸高剂量、中剂量和低剂量组。再灌注后,测定心肌组织ICAM-1表达、血脂,观察心肌病理形态学变化。结果与模型组比较,舒冠滴丸各剂量组心肌组织ICAM-1蛋白表达均降低(P<0.05),而辛伐他汀组与舒冠滴丸高剂量组相近;与模型组比较,舒冠滴丸高剂量和中剂量组可明显降低血清总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)水平(P<0.05),升高血清高密度脂蛋白胆固醇(HDL-C)水平(P<0.05),而辛伐他汀组血脂与舒冠滴丸高剂量组比较差异无统计学意义(P>0.05);舒冠滴丸高剂量组和辛伐他汀组缺血区心肌细胞排列基本整齐,心肌纤维断裂及坏死减少,空泡变性减轻,心肌间质水肿不明显,灶性出血及炎性细胞浸润减轻。结论舒冠滴丸可降低高脂血症大鼠缺血再灌注损伤心肌组织ICAM-1表达,减轻PMN浸润,调节血脂代谢紊乱。
Objective To investigate the effects of Shuguan dripping pill on the expression of intercellular adhesion molecule-1（ICAM-1） in hyperlipidemic rats with myocardial ischemia reperfusion injury and its mechanisms.Methods After hyperlipidemic rat models were established by oral administration of high lipid emulsion,the rats were divided into seven groups：Control group,sham group,model group,Shuguan dripping pill groups（high dosage,medium dosage,low dosage） and simvastatin group.After reperfusion,the blood samples were taken from the ventricles and assayed for blood lipid.Immunohistochemistry was used to determine ICAM-1 expression in myocardium.The myocardial pathological changes were observed.Results Compared with model group,the ICAM-1 expression in myocardium was decreased with the increase of Shuguan dripping pill dosage in Shuguan dripping pill groups（P〈0.05）.The levels of serum total cholesterol,triglyceride and low density lipoprotein cholesterol in high dosage and medium dosage Shuguan dripping pill groups were decreased and high density lipoprotein cholesterol was increased（P〈0.05）.In high dosage Shuguan dripping pill group and simvastatin group,myocardial fiber rupture and necrosis in ischemic area were lessened,and focal hemorrhage and inflammatory cell infiltration were decreased.Conclusion Shuguan dripping pill had protective effects on myocardial ischemia reperfusion injury in hyperlipidemic rats and its mechanisms might be related to decrease ICAM-1 expression in myocardium,inhibit neutrophil infiltration,and regulate blood lipid.
