摘要
目的:建立高效液相色谱-库仑阵列电化学法测定尿液中的神经内分泌瘤标志香草扁桃酸、高香草酸和5-羟吲哚乙酸,以及连用紫外检测法同时测定肌酐浓度的方法。方法:尿样经稀释离心后直接进样,采用50mmol/L磷酸二氢钠水溶液中分别含1%和40%的乙腈作为两相梯度分离,柱温35℃,主通道检测电压为160、260和360mV,紫外检测波长为210nm。结果:在浓度为0.1~1.5mg/L范围内,神经内分泌瘤标志的线性关系良好(r>0.9992),肌酐在30~450mg/L范围内线性关系良好(r=1.0000)。5-羟吲哚乙酸、香草扁桃酸、高香草酸和肌酐的平均回收率依次为96.9%、98.5%、101.2%和102.6%。四种物质的组内变异系数均<4%,组间变异系数均<6%。成人神经内分泌瘤患者的特征标志的值>正常对照值。结论:研究建立了快速准确同时检测随机尿样高香草酸、香草扁桃酸、5-羟吲哚乙酸和肌酐的方法,适用于临床神经内分泌瘤的诊断及其他神经精神等疾病的研究。
OBJECTIVE:To develop a HPLC-ECD method on simultaneous determination neuroendocrine tumor markers:5-hydroxyindoleacetic acid (5-HIAA),vanillylmandelic acid (VMA),homovanillic acid (HVA),and determination Cr using UV detector.METHODS:Samples were simply diluted and filtered before separation by binary gradient reversed phase chromatography.The mobile phase consisted of 50 mmol/L sodium dihydrogen phosphate with 1% and 40% acetonitrile as binary phases.The column temperature was 35 ℃.The dominant detection voltage was 160,260 and 360 mV.RESULTS:The calibration curve was linear in the range of 0.1~1.5 mg/L(r〉0.999 2) for the neuroendocrine tumor markers and in the range of 30~450 mg/L(r=1.000 0) for Cr.The methodological recovery of 5HIAA,VMA,HVA,and Cr was 96.9%,98.5%,101.2% and 102.6%,respectively.The intra-assay CV were less than 4% and inter-assay CV were 1ess than 6%.The values of major neuroendocrine tumor markers in adult patients were more than normal ranges.CONCLUSION:This HPLC method is accurate,sensitive,simple,and suitable for diagnosis of neuroendocrine tumor or other study such as neuropsychiatric disorders.
