摘要
通过定量分析大鼠坐骨神经的结构,观察糖尿病状态对外周神经的损害。将SD大鼠用四氧嘧啶腹腔注射诱发糖尿病模型后,非糖尿病大鼠及糖尿病大鼠随机分两组,饲养6个月后,在麻醉状态下,切取坐骨神经分别做光镜检查,包括:单神经外形频率分布,横截面细径分析,神经内膜毛细血管密度。电镜分析包括轴突,髓鞘,雪旺细胞的超微结构改变。结果表明,糖尿病大鼠组坐骨神经显示给旁脱髓鞘,节段性脱髓鞘,轴突萎缩,有髓纤维密度下降。糖尿病大鼠组糖代谢指标:血糖,山梨醇.糖基化血红蛋白水平显著高于非糖尿病对照组,神经内膜毛细血管密度两组无显著差异。提示:糖尿病状态可导致外周神经病变,病理机制可能是多因性的。
The aim of the study was to quantitatively evaluate effects of diabetic state on structure of sciatic nerve on alloxan-induced diabetic rats. Immediately after diabetic models were established, diabetic rats as well as non-diabetic rats were randomly divided into two groups. 6 months later, changes in glucose metuboic parameters including levels of blood glucose, HbA1c and sorbitol within red blood cell (RBC. S) morphologi of seiatic nerve, and capillary density of endonurium of all rats were investigated. Results demonstrated that: ① diabetic rats had greater abnormal structure than non- dlabetic control group in sciatic nerve including paranodal demyelination, myelin wrinkling, axon atrophy and myelinated fiber density. ② no sjgnificant differences were found in endonurium capillary density between the two groups. ③ the levels of blood glucose, HbA1c and RBC. S were higher in diabetic rats than in non-diabetic control group. This indicates that interplay of multiple factors in diabetic state can lead to peripheral neuropathy.
