基于连接酶检测反应的NPY基因多态与清远麻鸡繁殖性状的相关性研究

A Study on the Relativity between NPY Gene Polymorphism and Reproductive Traits of Qingyuan Partridge Chicken Based on Ligase Detection Reaction

摘要 为建立一个基于聚合酶链反应—连接酶检测反应的基因多态性检测系统,探讨清远麻鸡神经肽Y基因(NPY)的多态性,并分析其对清远麻鸡开产日龄、开产蛋重、开产体重、40周平均蛋重、50周平均蛋重以及52周产蛋数的遗传效应。应用PCR-LDR方法对NPY基因转录起始区4bp的缺失突变进行基因型分型。结果表明:基因型分型结果同直接测序分型结果一致。不同基因型个体间仅在开产体重上存在显著差异,BB型个体的开产体重显著低于AA和AB型个体。说明NPY基因多态影响清远麻鸡的开产体重。 The parallel typing system based on ligase detection reaction was established in the present study, and to detect the polymorphism ofNPY gene, as well as its genetic effect on age at first laying, egg weight at first laying, body weight at first laying, average egg weight at the age of 40 weeks, average egg weight at the age of 50 weeks, and total egg number at the age of 52 weeks in‘Qingyuan’partridge chicken. PCR-LDR technique was used to identify the genotypes of 4 bp deletion mutation in NPY transcription start region in ‘Qingyuan’partridge chickens. The genotyping results were coincidence with direct sequencing. The least square analysis showed that only body weight at first laying has significantly difference between different genotypes. BB genotype birds had significant lighter body weight at first laying than AA and AB genotype birds. There is a possible role ofNPY gene polymorphism in the body weight at first laying of‘Qingyuan’ partridge chicken.
