摘要
目的探讨心肺复苏后大鼠脑组织Caspase-3表达与细胞凋亡的关系。方法建立窒息联合冰氯化钾致大鼠心脏骤停/心肺复苏(CA/CPR)的动物模型,于各时相点断头留取脑组织,采用免疫组化的方法观察CPR后脑组织Caspase-3蛋白的表达,TUNEL检测神经细胞凋亡。结果对照组脑组织内仅有少量TUNEL阳性细胞;ROSC后3h出现表达阳性细胞,ROSC后6h表达开始增多,ROSC后12h进一步上升(P〈0.05),其后各组均处于高水平表达。Caspase-3表达均在ROSC后24h显著增加,直至ROSC后72h,与对照组比较差异均有统计学意义(P〈0.01),Speamaa等级相关检验Caspase-3表达与细胞凋亡明显相关(r=0.909)。结论CA/CPR后大鼠脑线粒体Caspase-3蛋白高表达可能是细胞凋亡增加的原因之一。
Objective To study the the relationship between expression of Caspase - 3 and apoptosis of neuron post cardiopulmonary resuscitation in rats. Methods Sprague Dawley rats cardiac arrest was induced by asphyxiation and ice - cold in 5 minutes of no circulation. 56 male Sprague Dawley rats were randomly divided into 7 groups : sham control group ( n = 8 ) ; after cardiopulmonary resuscitation the rats were allowed to reperfuse spontaneously for 3 h, 6 h, 12 h, 24 h, 48 h, 72 h, with each group of 8 rats. The rats were killed by decapitation and the brains were removed. The expression of Caspase - 3 and apoptosis of neuron in hippocampus of the model rats was observed by means of the immuhistochemistry and TUNEL respectively. Results Typical cell apoptosis was induced in neuron at 3 h and significant increase at 12 h post restoration of spontaneous circulation(ROSC) . Upregulation of Caspase - 3 expressions was maintained during 24 h post ROSC. Expression of Caspase - 3 in neuron was in accordance with the account of the apoptosis neuron. Conclusion Cardiac arrest and eardiopulmonary resuscitation induce the apeptosis of neuron, high expression of Caspase - 3 may one of the mechanism.
