摘要
目的:探讨黄芩醇提取物对牙龈卟啉单胞菌(Porphyromonas gingivalis,Pg)膜泡(extracellular vesicles,ECV)抑制牙周膜成纤维细胞(PDLFS)生物活性的影响。方法:采用细胞培养技术、细菌培养技术、3H-TdR掺入法,观察ECV对人牙周膜成纤维细胞DNA合成的抑制作用,以及不同浓度黄芩醇提取物对ECV这一生物活性的阻断作用。结果:细胞培养物中加入50μg/mL ECV时,人牙周膜成纤维细胞DNA的合成受到明显抑制,与空白对照组相比(P<0.05),当同时加入0.5、1、2μg/mL不同浓度黄芩醇提取物时其DNA合成量均有所增加,但以2μg/mL浓度组增加显著,与ECV组相比具有统计学意义(P<0.05)。结论:适宜浓度的黄芩提取物可有效地阻断ECV对HPDLFS生物活性的影响。
AIM:To investigate the effect of baicalin extract on DNA synthesis of human periodontal ligament fibroblasts(HPDLFS) under Porphyromonas gingivalis extracellular vesicles(ECV) stimulation.METHODS:Cultured HPDLFS were stimulated with 50μg/mL ECV or ECV plus different concentrations of baicalin for 36 h.HPDLFS with no stimulation served as the control.3H-TdR incorporation method was used to examine the synthesis of DNA in HPDLFS.RESULTS:DNA synthesis of HPDLFS in ECV group was significantly inhibited(P 0.05).Baicalin dose-dependently increased DNA synthesis of ECV-treated HPDLFS and 2 μg/mL baicalin showd statistically significant stimulating effects(P 0.05).CONCLUSION:Baicalin extract shows inhibitory effect on ECV,suggesting that it might be useful in the treatment of periodontal diseases.
