摘要
目的:研究白细胞介素-2基因转染人骨肉瘤细胞体外生物学特性.方法:应用逆转录病毒载体pDOR-neo将人的IL-2cDNA插入人骨肉瘤细胞,经G418筛选及PCR分析获得阳性细胞,并测定其上清液中IL-2含量.结果:PCR可从阳性细胞基因组中扩增出IL-2基因片段,并从其上清液中测得每24h105细胞IL-2活性为50U~800U.结论:逆转录病毒介导的IL-2CDNA可以在人骨肉瘤细胞系中获得有效表达,且转基因后的细胞较原野生型细胞体外生物学特性有明显差别.
AIM: To study in vitro biological characteristic ofhuman osteosarcoma cells transfected with IL-2 cDNA.METHODS: The human IL- 2 cDNA was inserted into the human osteosarcoma cells with the retroviral vector pDOR-neo.The human osteosarcoma cells secreting IL-2 were abtainedafter G418 screening and analysed by PCR. RESULTS: PCRwas used to identify the integration of the IL-2 gene in genomic DNA of G418-resistant human osteosarcoma cells. Theproduction of IL-2 was 50 U--800 U per 1×105 cells every 24h. CONCLUSION: IL-2 cDNA could be efficiently expressivein the tranduced human osteosarcoma cells and the resultsdemonstrated the in vitro biological characteristics of osteosarcoma cells transfected IL-2 gene were obviously different from the wild type ostosarcoma cells.
