摘要
目的:检测口腔鳞癌组织中3p14.2区D3S1300位点丢失情况,为新的抑癌基因定位提供依据。方法:应用聚合酶链反应(PCR)技术以D3S1300位点微卫星多态为遗传标记,检测口腔鳞癌组织中该位点的杂合性丢失(Losofheterozygosity,LOH)状况。结果:22例口腔鳞癌17例提供信息,7例出现杂合性丢失,LOH率为41%(7/17),有2例出现微卫星的不稳定性。结论:3p14.2区D3S1300位点杂合性丢失可能与口腔鳞癌的发生发展相关,提示该区域可能存在口腔鳞癌的抑癌基因。
Objective: To discuss the potential loss of tumor suppressor gene loci on chromosome 3p14.2 in oral squamous cell carcinoma (OSCC) and to provide meterials for locating new suppressor gene. Methods: We analyzed 22 pairing microdissected samples of normal tissues and OSCC from the same patients. LOH was detected by examing microsatellite polymorphic sequence in loci D3S1300 (3p14.2) by using PCR. Results: In 7 of 17 (41%), LOH was found at 3p14.2 (D3S1300). Microsatellite instability was found in two OSCC. Conclusion: This suggested that there was frequent loss on 3p14.2 in OSCC. There probably exist a suppressor gene which is important in oncogenesis and progression of OSCC. The mismatch repair gene which belongs to neither oncogene nor suppressor gene probably also plays an important role in OSCC.
