摘要
目的:研究离心沉淀集菌法的准确度。方法:以《中国药典》2005年版收录的离心沉淀集菌法为基础,对已知活菌数量的菌液或人工污染定量活菌的药品溶液进行离心操作,测定回收率。结果:3 000 r.min-1离心20或30 min,活菌细胞损失较大,不利于准确计数;500 r.min-1离心3 min,各试验菌的回收率可在60%以上。结论:离心沉淀集菌法不宜收录在微生物限度检查法中,在菌数测定等定量试验中应慎用或不用。
Objective: To study the accuracy of enrichment of microorganisms by centrifugation. Method: Based on the assay method of ChP 2005 edition, the recoveries of microorganisms were measured in the samples which it were contained or contaminated the viable known quantity. Results: Centrifuging at 3 000 r · min^-1 for 20 or 30 minutes, the viable microorganisms had a loss and cannot be counted out exactly. Centrifuging at 500 r · min^-1 for 3 minutes, the recoveries of microorganisms may be not less than 60%. Conclusion: Enrichment of microorganisms by centrifugation was unsuitable of being embodied in Microbial Limit Tests, and the method should be used cautiously and be disused at quantitative microbiological tests.
