摘要
扩增片段长度多态性(AmplifiedFragmentLengthPolymorphism)技术是一种兼具稳定性及多态检出率高的DNA指纹分析技术,具有广泛应用前景,但该技术费用高、操作较复杂.本文对AFLP技术从限制性内切酶水解、扩增条件、检测方法三方面作了一些改进,建立了一套稳定的和简便易行的实验条件,同时用这种改进的方法对家蚕基因组DNA的多态性进行了初步研究,得到了良好的DNA的多态性检测效果,不仅分辨力较高。
Amplified Fragment Length
Polymorphism (AFLP),a new DNA fingerprinting technique,is a reliable and powerful approach in
detecting polymorphism loci,but more expensive for a general laboratory and more complex in
perform.The approach of digestion of DNA,PCR conditions and detection method of PCR porducts
were improved in this paper.The results indicated that our method improves the efficiency of
detecting polymorphism loci.The method was applied to research silkworm genomic DNA and the
result shew that the electrophoresis bands were clear and stable.
