6个鹅种GH基因单核苷酸多态性检测及群体遗传分析

Identification and genetic analyses of single nucleotide polymorphism of GH gene in six domestic geese

摘要 确定鹅GH基因编码区是否存在单核苷酸多态性,并分析其在不同品种群体中的频率分布,为进一步探讨该基因与鹅生产性能关系而奠定基础。以太湖鹅、扬州鹅、豁眼鹅、浙东白鹅、皖西白鹅和四川白鹅6个鹅品种为试验材料,根据鸭、鹅GH基因序列设计了3对引物,分别扩增第3、4、5外显子,用PCR-SSCP方法进行单核苷酸多态性分析,并对不同品种群体进行群体遗传学分析。结果在引物2扩增片段上共检测到3个基因型(HH、HI和II),经过克隆测序比较发现2个单碱基突变,均为沉默突变,分别为编码区的T291C、G297A。统计结果发现3种基因型在6个鹅品种间分布存在极显著的差异(P<0.01)。所有品种均处于Hardy-Weinberg平衡状态。群体遗传分析表明豁眼鹅杂合度、有效等位基因数、多态信息含量最低,浙东白鹅最高;所有鹅种均处于中度多态。表明鹅GH基因不同品种中具有丰富的单核苷酸多态性,可以进一步作为候选基因来分析其与产肉性状的相关性。 The purpose is to identify single nucleotide polymorphism in goose GH gene,and to analyze the genetic distribution in different goose breeds,and then to provide finally a scientific basis for analyzing the correlation between the polymorphisms and production performance in goose.In this experiment,six goose breeds(Taihu goose,Yanzhou goose,Huoyan goose,Zhedong white goose,Wanxi white goose and Sichuan white goose)were used,three pairs of primers,basing on sequence of duck and goose GH gene,were designed,and then single nucleotide polymorphisms(SNPs) were detected by the technique of single strand conformation polymorphism(SSCP) and genetic polymorphism was analyzed in different breeds.Three genotypes(HH,HI and II) were detected in the PCR product of primer 2.By cloning and sequencing,two nucleotide variations of T291C,G297A were found,and they did not cause the change of amino acids.The result of χ2 analysis showed that the distribution of three genotypes was very different among breeds(P0.01).All breeds were in accordance with the Hardy-Weinberg equilibrium.The heterozygosity,effective number of alleles and polymorphism information content(PIC) were the lowest in Huoyan goose,but the highest in Zhedong white goose.All goose′ s PIC was in the middle level.The results showed that the GH gene had high polymorphisms in different goose breeds,and could be a candidate gene of analyzing the correlation between its polymorphism and production traits in goose.
