依达拉奉预防大鼠放射性口腔黏膜炎的初步研究 被引量:4

The effect of edaravone on the prevention of radiation-induced oral mucositis in rats

摘要 目的探讨依达拉奉对放射性口腔黏膜炎(ROM)的预防作用及其可能机制,为ROM提供新的防治方法。方法通过一次性照射SD大鼠舌背30GyX线,观察阴性对照组大鼠与用药组大鼠舌背黏膜的变化,其中用药组大鼠在接受照射前30min给予腹腔注射依达拉奉20mg/kg。照射后每天上午8时、下午4时共2次观察各组大鼠的活动度,称取体重;通过HE切片、免疫组化的方法,比较用药组和阴性对照组SD大鼠ROM的严重程度。结果经一次性照射30GyX线后,阴性对照组大鼠与用药组大鼠均出现ROM;用药组大鼠较阴性对照组大鼠的活动度较好,体重下降程度、舌部溃疡均较轻,白细胞介素6的表达较弱。结论依达拉奉可以显著减轻ROM的程度,放射前应用依达拉奉是一种预防ROM的有效方法。 Objective To investigate the effect of edaravone on the prevention of radiationinduced oral mucositis (ROM), and to explore the underlying mechanisms of ROM. Methods Tongues of Sprague-Dawley rats were exposed to 30 Gy radiation to establish the ROM model, Furthermore, the experimental group was given a intraperitoneal injection of edaravone 20 mg/kg before X-ray exposure, whereas SD rats without any treatment were served as negative control. Activity and weight of the SD rats in each group were recorded twice at 8am and 4pm every day, and differences between two groups were studied by HE staining assay and immunohistochemistry. Results Typical ulcerative change was observed in tongue mucosa in both groups after exposure to 30 Gy radiation once. Rats in the experimental group showed better activity and less seriousness in weight loss and ulcer degree. Interleukin-6 expression in tongue mucosa was also down-regulated compared to the control group. Conclusions Edaravone can mitigate ROM and showed potential role in ROM prevention.
