摘要
研究了聚二甲基硅氧烷(PDMS)膜生物反应器系统在长期封闭循环连续发酵中酵母细胞的适应性进化及发酵动力学。进行了3轮连续发酵实验,每轮发酵时间分别为495、512、440h。采集每轮发酵残液进行菌悬液涂平面培养基的菌株分离与筛选,并转移培养用于下一轮发酵。发酵实验中细胞和乙醇浓度均维持稳定。实验结果表明,筛选出的菌株在生产乙醇的能力上表现出一定优势,3轮发酵单位时间的细胞比产率分别为0.092、0.106、0.126g/g·h。对长期封闭循环连续发酵动力学行为进行了实验分析,获得了一些动力学实验基础参数。
The adaptative evolution of yeast cells and fermentation kinetics were investigated under long-term continuous close-circulation fermentation in a Polydimethylsiloxane(PDMS) membrane bioreactor system.Three rounds of continuous ferment experiments were carried out,with every round lasting 495,512,440 h,respectively.The living yeast cells were isolated and screened from the residual broth by spreading cells suspension on the culture medium.The selected strains were cultured and transferred to next ferment.The concentrations of cells and ethanol in broth were of a steady state during the ferment experiments.The experimental results showed that the selected yeast strains had dominant character to some extent in ethanol production.The productivities of ethanol -cells per hour were 0.092,0.106,0.126 g/g·h for every round of ferment,respectively.The kinetics of long-term continuous close-circulation fermentation was analyzed and the experimental kinetics parameters were obtained.
