摘要
目的:优选丹芪偏瘫胶囊提取液(简称DQ液)的提取工艺;通过比较不同醇沉处理的DQ液指纹图谱的差异性,客观反映醇沉浓度对药物总体成分的影响。方法:以药物醇沉浓度为考察因素,以水溶性有效成分为指标,采用单因素考察设计,采用反相高效液相色谱法,色谱柱:Agilent Eclipse Plus柱,C18(4.6mm×250mm,5.0μm)。流动相乙腈-水-冰乙酸,梯度洗脱。柱温:35℃,进样量:10μL,检测波长:254nm。对不同浓度醇沉后DQ液进行HPLC测定,并采用中药色谱指纹图谱相似度评价软件进行比较。用指纹图谱研究方法对DQ液作醇沉浓度的优选。结果:丹芪偏瘫胶囊的最佳醇沉浓度为65%。结论:建立了DQ液的HPLC指纹图谱,可用于其生产过程中的质量控制。不同浓度醇沉液的指纹图谱差异较明显,各指标成分在65%醇沉时含量最高。
Objective:To optimize the extraction of Danqi hemiplegia capsule (Referred to as DQ),by comparing differences of DQ fingerprints of the different alcohol precipitation,objectively reflect the concentration of alcohol precipitation impact on the overall medicines.Methods:The concentration of alcohol precipitation as study factor,Water-soluble active ingredients as an indicator,apply of single-factor study and the RP-HPLC,C18 column (4. 6mm×250mm,5μm) was used with the mobile phase containing acetonitrile-water-acetic acid for gradient elution.The injection volume was 10 L,The column tempreture was 35℃,Wavelength detection at 254nm.Results:Optimal concentration of alcohol precipitation of the extraction of Danqi hemiplegia capsule was 65%. Conclusion:Establish the HPLC fingerprint of Danqi hemiplegia capsule ,which can be used in its quality control of production process.There are obvious different among the fingerprints of the different alcohol precipitation,The active ingredients reach their peak when concentration of alcohol precipitation is 65%.
