摘要
建立了一种快速测定红曲米中桔霉素含量的高效液相色谱方法。比较了不同萃取剂、提取温度、超声波处理时间对桔霉素提取效果的影响,使用Shimpack HRC-ODS色谱柱(4.6 mm×150 mm,5μm),以甲醇-水(50:50,V/V)为流动相,采用二极管阵列检测器(检测波长254 nm)对桔霉素进行测定。结果表明,采用甲苯-乙酸乙酯-甲酸(7:3:1,V/V/V)在50℃下提取,超声波处理1 h,可将桔霉素提取完全,在2~24μg/mL范围内桔霉素含量与峰面积成良好的线性关系,相关系数R^2=0.997 3,精密度和稳定性试验相对标准偏差均小于5%,加样回收率达到95%,最低检测限为0.1 mg/L。该测定方法简单、准确、精密度高、重现性好。
A HPLC method for rapid determination of Citrinin in Monascus was developed.The effects of different factors including extracting solvent,extraction temperature and ultrasonic extraction time on the extraction of citrinin were compared.Citrinin was separated by mobile phase of methanol -water on Shimpack HRC - ODS column, and detected at 254 nm by photodiode array detection.Results:Citrinin can be extracted completely by Tolueneethyl acetate -formic acid(7:3:1,V/V/V) at 50℃,with 1h ultrasonic processing.The extracted citrinin presents a good linear relation between concentration and peak area within a range of 2~24μg/mL(R^2=0.997 3).The relative standard deviations(RSD) of reproducibility and stability experiments are lower than 5%,the average recovery rate of citrinin is 95%and the detecting limit is 0.1 mg/L,proving that the method is a simple and accurate method with high reproducibility.
