摘要
目的:建立以反相高效液相色谱法测定清开灵3种不同药物剂型中绿原酸含量的方法。方法:色谱柱为Sino Chrom ODS-AP C18(250mm×4.6mm,5μm),流动相为甲醇-水-冰醋酸(25∶75∶0.5),流速为1.0mL·min-1,检测波长为327nm,柱温为30℃。结果:绿原酸的检测浓度在3.81～152.50μg·mL-1范围内与峰面积积分值呈良好线性关系(r=0.9998);清开灵胶囊、颗粒、滴丸的平均回收率分别为99.03%(RSD=2.02%)、99.35%(RSD=1.16%)、98.11%(RSD=1.51%)。结论:本法灵敏、简便、准确,可用于清开灵3种不同剂型的绿原酸含量测定和质量控制。
OBJECTIVE： To establish RP-HPLC method for the content determination of chlorogenic acid in 3 kinds of dosage forms of Qingkailing preparation. METHODS： SinoChrom ODS-AP C18（250 mm×4.6 mm, 5 μm） column was adopted and the mobile phase consisted of methnol-water-acetic acid （25 ∶ 75 ∶ 0.5） at the flow rate of 1.0 mL·min-1. Column temperature was set at 30 ℃ and the detection wavelength was set at 327 nm. RESULTS： The linear range of chlorogenic acid was 3.81～152.50 μg·mL-1（r=0.999 8）, and the average recoveries of the capsules, granules and dripping pills were 99.03%（RSD=2.02%）, 99.35%（RSD=1.16%）,98.11%（RSD=1.51%）. CONCLUSION： The method is sensitive, simple and accurate for the quality control and content determination of 3 kinds of Qingkailing preparations.
