摘要
目的:探讨非小细胞肺癌中DNA修复酶O6-甲基鸟嘌呤-DNA甲基转移酶基因的过甲基化状况及表达的关系。方法:采用甲基化特异性PCR及免疫组织化学法检测96例经病理确诊的非小细胞肺癌中DNA修复酶O6-甲基鸟嘌呤-DNA甲基转移酶基因及蛋白表达。结果:96例非小细胞肺癌组织DNA修复酶O6-甲基鸟嘌呤-DNA甲基转移酶基因过甲基化率36.5%,DNA修复酶O6-甲基鸟嘌呤-DNA甲基转移酶蛋白的阳性表达率为33.3%,DNA修复酶O6-甲基鸟嘌呤-DNA甲基转移酶基因过甲基化与蛋白的表达呈负相关(Rs=-0.536,P<0.05)。结论:在非小细胞肺癌中,DNA修复酶O6-甲基鸟嘌呤-DNA甲基转移酶基因过甲基化导致蛋白表达的降低,可能与肺癌的发生、发展相关。
Objective To investigate the relationship between the promoter hypermethylation and expression of O6-methylguanine DNA methyltransferase gene in non-small cell lung cancer.Methods Methylation specific PCR and immunohistochemical methods were used to detect O6-methylguanine DNA methyltransferase gene and protein in 96 cases of pathologically confirmed non-small cell lung cancer.Results The rate of the promoter hypermethylation of O6-methylguanine DNA methyltransferase gene was 36.5%,and the rate of the expression of O6-methylguanine DNA methyltransferase gene was 33.3% in non-small cell lung cancer.The promoter hypermethylation was negatively correlated to the expression of O6-methylguanine DNA methyltransferase in non-small cell lung cancer(Rs=-0.536,P0.05).Conclusion The promoter hypermethylation of O6-methylguanine DNA methyltransferase gene may lead to the reduction of the protein,and it may be involved in lung cancer.
