摘要
目的:建立以柱前衍生化高效液相色谱-荧光检测法测定高血压患者尿液中22种氨基酸浓度的方法。方法:衍生化试剂为洗丹脱磺,流酰速氯为,色1.0谱m柱L·为mKinr-o1m,发asi射lC波18,长流为动5相08为nm甲,醇激与发3波0长mm为o3l·3L0-n1m醋,酸柱钠温缓为冲室溶温液,(进p样H量7.2为)-5甲0μ醇L-。四结氢果呋:2喃2(种84氨:1基5酸:1)检,测进浓行度梯度分别在5~200μmol·L-1范围内线性关系良好(r>0.9900);平均回收率为86.1%~109.0%;进样精密度RSD为0.84%~3.1%(n=6)。结论:本方法可同时测定22种氨基酸的浓度。
OBJECTIVE:To establish an HPLC method for the determination of 22 kinds of free amino acids in urine of hypertensive patients.METHODS:The determination was performed on a Kromasil C18 column with mobile phases of methanol and 30 mmol·L^-1 sodium acetate buffer(pH 7.2)-methanol-tetrahydrofuran(84:15:1)(gradient elution).Flow rate was set at 1.0 mL·min^-1.The fluorescence excitation and emission wavelengths were 330 and 508 nm,respectively.Column temperature was room temperature and injection volume was 50 μL.Dansyl chloride was used as derivatization agent.RESULTS:The linear range of free amino acids were 5~200 μmol·L^-1(r〉0.990 0) with an average recovery ranged from 86.1% to 109.0%.The injection RSD were within 0.84%~3.1%(n=6).CONCLUSIONS:The method is successfully applied for simultaneous determination of 22 kinds of free amino acids.
