摘要
研究优化了大麻基因组原位杂交(Genomic in situ hybridization,GISH)技术,并成功应用于外源遗传物质的鉴定,为进一步分析大麻的基因组结构、特别是研究大麻的性别分化奠定基础。切口平移法生物素(Biotin)标记探针,探针与大麻染色体在80℃共变性6分钟,可以将亚麻基因组探针成功的与大麻染色体杂交,当标记基因组总DNA与封阻基因组总DNA的浓度比例为1:200时,能有效区分大麻与亚麻的基因组。
Genomic in situ hybridization(GISH)technique,the DNA of flax as probe,was improved in hemp in the study,and it is successful for identifying extraneous DNA.It showed that the modified procedures could be further used to study gene structure of hemp,especially to study the sex differentiation of the hemp.With probe DNA labeled by nick translation,and co-denaturation of chromosome DNA for 6 min at 80℃,the DNA of flax was successfully localizated by GISH method,when the ratio of marked genome DNA and the blocked genome DNA was 1:200,the genomes of flax and hemp could be separated effectively.
