Expression of Fas Antigen and Fas- Ligand in Skin Lesions of Patients with Lupus Erythematosus

摘要 目的研究Fas抗原及Fas配体(Fas L)在红斑狼疮皮损中的表达情况。方法应用免疫组化技术检测25例系统性红斑狼疮(SLE)及14例盘状红斑狼疮(DLE)不同病程的皮损中Fas抗原及Fas L的表达。结果SLE及DLE早期皮损角朊细胞Fas抗原表达强度显著高于正常皮肤(P<0 01) ,与病程呈负相关(P<0 05) ,且真皮中单一核细胞亦有Fas抗原表达 ;红斑狼疮患者皮损及正常皮肤的角朊细胞均不表达Fas L ,仅在早期皮损真皮内单一核细胞表达Fas L。结论Fas系统在角朊细胞及真皮异常表达可能与红斑狼疮皮损发生。 Objective To investigate the expression of Fas antigen and Fas ligand (Fas L) protein in the skin lesions of patients with lupus erythematosus Methods An immunohistochemical technique was performed for detecting Fas antigen and Fas L expression in the skin lesions of 25 cases of systemic lupus erythematosus (SLE) and 14 cases of discoid lupus erythematosus (DLE) Results The positive Fas stains were observed in keratinocytes of early skin lesions of both DLE and SLE, and were significantly stronger than those of normal skin (P< 0 01), and the expression was negatively related to the course of the disease (P< 0 05) Positive Fas stains were also found in mononuclear cells(MNCs) of dermis However,Fas L was not expressed in keratinocytes in both skin lesions and normal skin of DLE and SLE, only existed in MNCs of dermis in early stage lesions Conclusion The abnormal expression of Fas and Fas L in keratinocytes and dermal MNCs might be related to the occurrence and development of lupus skin lesions in both DLE and SLE
