摘要
为了筛选耐盐、抗真菌的几丁质酶高酶活菌株用于生物防治,从黄海连云港沿岸、运城盐湖、青海湖及茶卡盐湖采集泥样16份,以胶体几丁质为唯一碳源和氮源,分离到产几丁质酶的细菌174株,放线菌82株,真菌60株.用对峙法测定产几丁质酶菌株对植物病原真菌尖孢镰刀菌的抑制作用,有24株细菌、7株放线菌及26株真菌抑菌区宽度≥6 mm.用胶体几丁质和DNS法测定细菌几丁质酶活力,有8株细菌酶活力≥1.67×10-9mol/s.选取抑菌区≥6 mm或酶活力≥1.11×10-9mol/s的38株细菌做耐盐性实验,有13株细菌属于耐盐菌,25株细菌属于极端耐盐菌.
To isolate some chitinase-producing microorganisms with anti-fungal strains and salt tolerance for the biological-control,16 soil samples,collected from the Yellow Sea coast of Lianyungang,Yuncheng salt lake,Qinghai Lake and Chaka Salt Lake,China,used as maerix,174 bacterias strains using chitin as carbon and nitrogen source, 82 actinomyces strains and 60 fungis strains were isolated.The inhibition of the samples to the phytopathogenic fungi was also investigated.The results showed that 24 bacterias strains,7 actinomyces strains and 26 fungis strains have significantly inhibitive effect on the growth of Fusarium oxysporum.The chitinase activity was determined by the method of DNS(3,5-dinitrosalicylic acid).Chitinase activity of 8 bacterial strains was higher than 1.67×10^-9mol/s.38 bacteria strains with inhibiting board more than 6 mm and chitinase activity≥1.11×10^-9 mol/s were selected for salt-tolerance experiment.The results indicated that 13 strains are of salt-tolerant bacterium and 25 strains are of extreme salt-tolerant bacterium.
