摘要 目的研究雌激素受体β(ERβ)对人牙周膜成纤维(HPLF)细胞骨钙素(OCN)含量的影响。方法采用基因干涉方法抑制了HPLF细胞中ERβ基因的表达,用雌激素干预ERβ抑制的和未抑制的HPLF细胞,研究细胞骨钙素含量的变化情况。结果测序鉴定重组质粒后,蛋白质印迹法及RT-PCR结果显示ERβsiRNA载体可特异地抑制HPLF细胞中ERβ的表达。经雌激素干预后,HPLF细胞OCN含量明显高于对照组(P<0.01),但ERβsiRNA载体稳定转染的HPLF细胞OCN含量与对照组相比无统计学意义。结论雌激素可能通过ERβ亚基促进人牙周膜成纤维细胞的骨钙素含量增加。 Objective To investigate the effect of estrogen receptor beta(ERβ) on osteocalcin production in human periodontal ligament fibroblast(HPLF) cells.Methods A short interfering RNA(siRNA) technique was used to inhibit ERβ expression in HPLF cells cultured with 17β-estradiol at a saturating concentration of 10-7mol/L.The amount of osteocalcin was determined with an osteocalcin ELISA kit.Results RT-PCR and Western blot showed that ERβ mRNA vector could specifically inhibit the expression of ERβ in HPLF cells.The osteocalcin level in HPLF cells was significantly higher in estradiol treatment group than in control group(P0.01).However,no significant difference was observed in ERβ siRNA vector-transfected osteocalcin in HPLF cells between the two groups(P0.05).Conclusion Estrogen may stimulate osteocalcin production in HPLF cells mainly via ERβ.
