摘要
为增强肝癌肿瘤浸润淋巴细胞(TIL)的趋肝性,制备了半乳糖基抗CD3单克隆抗体-TIL(Gal-antiCD3McAb-TIL),将其与肝细胞共同孵育,在倒置显微镜下观察两者通过无唾液酸糖蛋白受体介导的作用情况。测定了半乳糖基抗CD3单抗(Gal-antiCD3McAb)的糖密度以及Gal-antiCD3McAb与TIL的结合率。结果发现,Gal-antiCD3McAb-TIL与肝细胞有明显粘附现象。Gal-antiCD3McAb的糖密度为62.18,Gal-antiCD3McAb与TIL的结合率为97.9%。结果表明,Gal-antiCD3McAb-TIL在体外有良好的趋肝性,62.18的糖密度和97.9%的结合率保证了TIL的有效利用。
This study was undertaken to enhance the hepatic targeting tendency of tumor infiltrating lymphocytes (TILs) and hence to lower the recurrence rate of primary liver cancer after hepatectomy. Galactosyl anti CD 3McAb TILs were prepared and then were incubated together with hepatocytes. Their interaction through asialoglycoprotein receptor mediated mechanism was observed under the inverted phase contrast microscope. The carbohydrate density of galactosyl anti CD 3McAb and the combining rate of galactosyl anti CD 3McAb with TILs were measured. The results revealed that galactosyl anti CD 3 McAb TIL obviously were adhered to hepatocytes. The carbohydrate density of galactosyl anti CD 3McAb was 62.18, and the combining rate of galactosyl anti CD 3McAb with TILs was 97.9%. The results suggested that in vitro hepatic targeting tendency of galactosyl anti CD 3McAb TILs was satisfactory, and that the carbohydrate density of 62.18 and the combining rate of 97.9% ensured the effective use of TILs.
