摘要
研究丙谷胺(PGL)及其与生长抑素(SST)联合应用对人胃癌细胞株MKN45裸鼠移植瘤的抑制作用。方法建立人胃癌细胞株MKN45移植瘤的动物模型,观察PGL(500mg/kg体重)、SST(200μg/kg体重)及两者联合应用对胃癌移植瘤的面积、重量、胃癌细胞DNA的影响。结果PGL组、PGL+SST组移植瘤的面积明显缩小,肿瘤重量明显减轻,胃癌细胞DNA指数、平均DNA含量及S期细胞比例明显降低(P<0.05),PGL+SST组更为显著。结论PGL能抑制胃癌细胞株MKN45移植瘤的生长,SST能明显增强PGL对移植瘤的抑制作用。
Objective To study the inhibitory effect of proglumide (PGL) and the combination of PGLand sandostatin (SST) on exnotransplanted human gastric cancer cell line MKN45 in nude mice. Method Theanimal models of xenotransplanted human gastric cancer cell line MKN45 were established in nude mice. Theresults of PGL (500 mg/kg), SST (200 μg/kg), PGL+SST combination in the tumor size, weight, DNAof exnotransplanted gastric cancer cells were observed. Result In PGL. and PGL+SST groups, the tumorsize is much smaller, the tumor weight much lighter, DNA index, average DNA content and S phase fractionof gastric cancer cells lower than in control group (P<0.05). Conclusion PGL can inhibit the growth ofxenotransplanted gastric cancer cell line MKN45. Short-time use of SST can significantly enhance the inhibitory effect of PGL.
