摘要
目的探讨SLE人群和正常对照干扰素调节因子5(IRF5)基因表达的差异。方法收集92例活动期的SLE病例与88名正常对照,提取外周血总RNA并反转录为cDNA,采用实时荧光定量聚合酶链反应扩增IRF5基因,获得扩增体系的Ct值后,用公式2-⊿⊿Ct计算SLE组和正常对照组的IRF5的mRNA表达水平。SLE患者抗ds-DNA抗体检测采用免疫印迹法。结果利用实时荧光定量聚合酶链反应扩增得到了预期大小的IRF5基因片段;SLE组、正常对照组外周血IRF5mRNA的表达水平分别为0.232±0.076、0.081±0.040,SLE组IRF mRNA表达水平升高(P<0.05)。SLE患者抗ds-DNA抗体阳性组IRF5基因表达水平有增高的趋势,但与阴性组相比没有差异。结论系统性红斑狼疮SLE患者外周血IRF5表达异常,可能在系统性红斑狼疮组发病中起一定作用。
Objective To investigate whether the expression of IRF5 is abnormal in systemic lupus erythematosus patients.Methods The clinical data of 92 systemic lupus erythematosus patients and 88 normal controls were collected.Peripheral blood was drawn from every subject in the morning.The total RNA of peripheral blood was extracted and transcripted to cDNA.SYBR green I real-time quantitative polymerase chain reaction was used to amplify IRF5 gene.Ct of every subject was obtained from real-time quantitative polymerase chain reaction system.The mRNA expression was indicated as the value of 2^-⊿⊿Ct.IRF5 mRNA expression in systemic lupus erythematosus patients and normal controls was compared.ds-DNA antibody was detected by immunoblotting method.Results The expected fragments of IRF5 gene were amplified by real-time quantitative polymerase chain reaction.The levels of IRF5 mRNA in systemic lupus erythematosus patients and normal controls were 0.232±0.07 and 0.081±0.040 respectively(P〈0.05).The levels of IRF5 mRNA in SLE ds-DNA antibody positive group tend to rise,but the difference in ds-DNA antibody positive and negative group was not significant.Conclusion Abnormal IRF5 mRNA expression is found in the peripheral blood of systemic lupus erythematosus patients.IRF5 may play roles in the pathogenesis of systemic lupus erythematosus.
