摘要
目的:探讨Toll样受体4(Toll-like receptor4,TLR4)、转录因子激活蛋白-1(activating protein-1,AP-1)(通常是c-Jun和c-Fos的异源二聚体)及核转录因子-κB(nuclear factor-kappa B,NF-κB)在非小细胞肺癌(non-small cell lung cancer,NSCLC)中的表达情况及其临床意义。方法:采用免疫组织化学SP法检测54例NSCLC组织及54例癌旁正常肺组织中TLR4、c-Jun、c-Fos及NF-κB蛋白的表达,并分析其与患者临床病理特征的关系。结果:TLR4、c-Jun、c-Fos及NF-κB蛋白的表达在NSCLC组织中高于癌旁正常肺组织,且差异具有统计学意义(P<0.05)。TLR4、AP-1及NF-κB蛋白的表达分别与NSCLC组织的分化程度、有无淋巴结转移及组织类型有关。在NSCLC组织中,TLR4与c-Jun、c-Fos及NF-κB的蛋白表达水平之间呈正相关。结论:TLR4、AP-1及NF-κB的高表达可能与NSCLC的发生发展有着密切关系,对其检测有助于判断肺癌的恶性程度及生物学行为。
Objective:To investigate the expression of Toll-like receptor 4(TLR4),activating protein-1(AP-1,heterodimer of c-Jun and c-Fos) and nuclear factor-kappa B(NF-κB) in human normal lung tissues and non-small cell lung cancer(NSCLC) tissues,and to explore their clinical significance.Methods:Immunohistochemical SP method was used to detect the expression of TLR4,c-Jun,c-Fos,and NF-κB in 54 cases of NSCLC and 54 cases of para-cancerous normal lung tissues.The correlation of TLR4,AP-1,and NF-κB with clinical pathologic features of lung cancer was analyzed.Results:The expressions of TLR4,c-Jun,c-Fos,and NF-κB protein were significantly higher in NSCLC tissues than those in adjacent normal lung tissues.The difference was significant(P0.05).Expression of TLR4,AP-1 and NF-κB correlated with tumor differentiation,lymph node metastasis and histological classification.In addition,the expression of TLR4 was positively associated with the expression of c-Jun,c-Fos,and NF-κB,respectively.Conclusion:The over-expression of TLR4,AP-1 and NF-κB in lung cancer cells had close relationship with the tumorigenesis and development of NSCLC.Therefore detection of TLR4,AP-1 and NF-κB may help to estimate the malignant degree and biological behavior of NSCLC.
