摘要
目的探讨Hedgehog信号通路相关基因在人脑胶质瘤干细胞及脑肿瘤组织中的表达及其意义。方法用半定量逆转录-聚合酶链反应(RT-PCR)法检测U251和D341恶性脑胶质瘤干细胞中SHH、Ptch、Smo mRNA及23例脑胶质瘤组织中SHH和Smo mRNA的表达。结果 SHH mRNA在U251细胞和胶质瘤组织中的表达分别显著高于其在D341细胞和瘤周组织中表达(P<0.05);Ptch mRNA、Smo mRNA在两个细胞系细胞中的表达及Smo mRNA在胶质瘤组织和瘤周组织中的表达无显著性差异(P>0.05)。结论本结果提示,Hedgehog信号通路的异常启动参与了脑胶质瘤的病理发生、发展过程。
Objective To explore the expressions of Hedgehog(HH) signal pathway genes including SHH,Ptch and Smo mRNA in human brain glioma stem cells and the human brain glioma tissues and their meaning.Methods The expressions of SHH,Ptch,and Smo mRNA in brain glioma stem cells U251 and D341 cell lines and the expressions SHH and Smo mRNA in 23 specimens of brain glioma tissues and the peritumorous tissues were determined by semi-quantitative RT-PCR.Results The expression level of SHH in U251 cell line and brain glioma tissues were significantly higher than those in U341 cell line and the peritumorous tissues respectively(P〈0.05).There was insignificant differences in the expression of Ptch and Smo RNA between both U251 and U341 cell lines(P〉0.05).There was insignificant differences in the expression of Smo RNA between the glioma tissues and the peritumorous tissues(P〉0.05).Conclusion It is suggested that the abnormal activation of HH signal pathway may take part in the pathogenesis and development of human brain glioma.
