摘要
目的研究含RGD序列合成肽对人喉癌Hep-2细胞株增殖能力的影响,并对其可能机制进行初步探讨。方法化学合成RGD肽,免疫荧光技术检测RGD肽与Hep-2细胞结合能力;MTT法检测不同浓度RGD肽对喉癌细胞增殖能力的影响;免疫细胞荧光技术检测RGD肽处理前后喉癌细胞整合素αv蛋白表达。结果 RGD肽与喉癌Hep-2细胞有特异性结合作用,经RGD肽处理后的喉癌细胞增殖能力显著降低,整合素αv表达明显降低。结论作为整合素αv受体拮抗剂,RGD肽对喉癌细胞的增殖能力有明显的抑制作用,为喉肿瘤基因治疗方面提供一个新靶点。
Objective To investigate the inhibitory effects of RGD peptide on proliferation of laryngeal cancer cells.Methods The peptide contained RGD sequence was synthesized and its effect of binding to laryngeal cancer cell was detected by immunofluorescent technology.After treatment with the RGD peptide,the expression of integrin alpha-v in laryngeal cancer cells was detected by immunofluorescence,the cell proliferation was assessed by MTT assay.Results RGD peptide could specifically bind to laryngeal cancer cells.The proliferation of the cells treated with RGD peptide was significantly decreased.The expression of integrin alpha-v in laryngeal cancer cells treated by RGD peptide was significantly decreased.Conclusion As an alpha-v integrin antagonist,RGD peptide can inhibit the proliferation of laryngeal cancer cells.RGD peptide may become an ideal therapeutic target for genic therapy of laryngeal tumors.
