摘要
目的:将纯化的Survivin蛋白作为诱饵,从噬菌体肽库中筛选出结合肽。方法:用纯化的Sur-vivin蛋白包被35 mm的塑料培养皿,加入随机12肽的噬菌体肽库,用靶分子溶液竞争性洗脱结合的噬菌体,通过测定噬菌体滴度检测Survivin蛋白的吸附、富集效果,筛选阳性噬菌斑,制作DNA测序模板进行测序。结果:经3轮筛选,噬菌体显示有良好的富集效果。从第3轮筛选产物中挑取8个蓝色噬菌斑(阳性克隆)进行测序,DNA测序结果表明,具有一致性序列GIANNFFTLKIS。结论:从噬菌体随机肽库中,筛选到能与Survivin蛋白特异性结合的一段短肽,为Survivin蛋白的进一步研究奠定基础。
Objective:To screening Binding-peptide from phage peptide library utilizing the decoy of the purified Survivin proteinum.Methods: By using a 35mm plastic culture capsule invested by purified Survivin proteinum,dodeca-peptid of phage peptide library was added.To removed the unconjugated and competing eluted the conjugated bacterial invader by target molecule liquid,the adsorbing and enriching effect were evaluated by the titre of bacterial invader.Positive negative colony was screeninged and then was made to DNA sequencing template.Results:After three times screening,bacterial invader shows the enriching effect.From the third time,eight blue bacterial invader(positive clone) was choosed for DNA sequencing,they had common sequence: GIANNFFTLKIS.Conclusion:Binding-peptidewhich can be screened from phage peptide library by the decoy of the purified Survivin proteinum,and is the foundation of investigating the Survivin proteinum.
