航天诱变混菌固态发酵玉米秸秆生产单细胞蛋白条件的优化 被引量:4

Optimization on the condition of space-flight mutantion mix-strains solid-state fermenting corn stalk for producing single cell protein

摘要 通过正交试验对啤酒酵母YB-6的接种时间、接种量和黑曲霉ZM-8与啤酒酵母YB-6共生发酵时间等因素的参数进行了优化。结果表明,啤酒酵母YB-6的接种时间(A)、接种量(B)和共生发酵时间(C)对CMC酶、FPU酶和SCP含量的影响大小顺序均为A>C>B,且产CMC酶和SCP的最优组合为A3C2B2,即酵母菌的接种时间为黑曲霉ZM-8发酵24h后、共生发酵时间为48h、接种量为10%;产FPU酶的最优组合为A4C3B3,即酵母菌的接种时间为黑曲霉ZM-8发酵36h后、共生发酵时间为60h、接种量为15%。最后,综合各因素将发酵条件的最优组合确定为A3C2B2。优化后的FPU酶、CMC酶和SCP含量分别为6.83、28.12U/g和26.73%,比优化前测定各指标值均有不同程度的提高。 The effecting factors of the mix-strains solid-state fermenting corn stalk for produing cellulase activities and single cell protein including the inoculation time (A)and inoculation amount (B)of Saccharomyces cerevisiae YB-6, and the time of Saccharomyces cerevisiae YB-6 and Aspergillus niger ZM-8 co-fermentation (C) were studied in the trial.This article optimized the parameters of those factors by orthogonal experiment.The result indicated that the influnce order of those factors to CMC,FPU and the content of SCP was A〉C〉B,and the optimal combination of producing CMC and SCP was A3C2B2,the inoculation time of Saccharomyces cerevisiae YB-6 was after 24 hours of Aspergillus niger ZM-8 fermentation,the inoculation amount of it was 10 % and the time of co-fermentation was 48 hours,the optimal combination of producing FPU was A4C3B3 the inoculation time of Saccharomyces cerevisiae YB-6 was after 36 hours of Aspergillus niger ZM-8 fermentation,the inoculation amount of it was 15 % and the time of co-fermentation was 60 hours.It concluded that the combination was A3C2B2.Under the optimized conditions,the activities of FPU,CMC and the content of SCP in the fermentation medium were respectively 6.83,28.12 U/g and 26.73 %,which was higher than before.
