摘要
目的检测解整合素-金属蛋白酶33(ADAM33)基因单核苷酸多态性与山东汉族人群慢性阻塞性肺疾病(COPD)的关系。方法采用PCR-RFLP对363例COPD患者和372名健康人ADAM33基因T1位点多态性进行分型,对等位基因及基因型分布进行统计学分析。结果 ADAM33基因T1位点3种基因型AA、GA、GG基因型频率在病例组中分别为0.8265、0.1680和0.0055,在对照组中分别为0.8763、0.1156和0.0081,A和G等位基因型的频率在病例组中分别为0.9105和0.0895,在对照组分别为0.9341和0.0659,该位点基因型和等位基因型分布在病例组与对照组P值分别为0.117,0.090,差异均无统计学意义。结论 ADAM33基因T1位点多态性与山东汉族人群COPD发生无明显相关性。
OBJECTIVE To investigate the relationship between the polymorphism of T1 locus allele in a disintegrin and metalloprotease 33(ADAM33) gene and chronic obstructive pulmonary disease(COPD) in the Chinese Han population from Shandong Province.METHODS To determine the polymorphism of T1 locus allele in ADAM33 gene,we used polymerase chain reaction(PCR) based restriction fragment length polymorphism(RFLP) technique in 363 COPD subjects and 372 healthy controls.RESULTS The frequencies of genotypes AA,GA and GG at polymorphism sites T1 were 0.8265,0.1680 and 0.0055,respectively in COPD group,while 0.8763,0.1156 and 0.0081 in control group.Besides,the A type allele frequencies were 0.9105 and 0.9341 in patients and controls respectively,and the G type allele frequencies were 0.0895 and 0.0659 in COPD patients and healthy controls.There was no significant difference between genotype frequencies(P=0.117) or allele frequencies(P=0.090).CONCLUSION The polymorphism of T1 locus allele in ADAM33 gene have none association with the susceptibility to COPD in Chinese Han population from Shandong Province.
