摘要
通过对红皮云杉胚性愈伤组织保持与增殖阶段各影响因子的筛选与分析,确定出适宜该阶段培养的培养基为附加5g·L-1肌醇、0.21mg·L-1BA、0.20mg·L-1KT、1.85mg·L-1NAA、20g·L-1蔗糖、450mg·L-1L-谷氨酰胺、750mg·L-1水解酪蛋白、2g·L-1凝胶(Gelrite)的改良RJW基本培养基,在22℃条件下暗培养。结果表明:以固体/滤纸交替培养方式的增殖率、增殖量最高,胚性愈伤组织诱导率与增殖率、增殖量之间存在正相关关系。随着继代次数的增加,增殖率和增殖量却逐渐下降,不同的细胞系下降的速率也不相同。
A study was conducted to screen the influence factors on the maintenance and proliferation of Picea koraiensis embryogenic callus by solid and filter paper alternative culture. The optimal culture medium was proved to be the improved basic medium RJW supplemented with 0.21 mg·L-1BA,0.20 mg·L-1KT,1.85 mg·L-1NAA,20 mg·L-1 sucrose,450 mg·L-1 L-glutamine,750 mg·L-1 casein hydrolysate,and 2 g·L-1 gelrite. Culture was carried out at 22 degrees C under dark condition. Results showed that the proliferation rate and proliferation quantity of embryogenic callus for the solid and filter paper alternative culture were highest,and induction rate of embryogenic callus was positive correlated with proliferation rate and proliferation quantity. The proliferation frequency and proliferation quantity decreased gradually with the increase of subculture times,and different cell lines showed different descending rates.
