摘要
目的:通过对感染乙肝病毒的流产孕妇外周血HBV-DNA含量检测和其淋巴细胞染色体着丝粒点改变研究分析,探讨孕妇自然流产原因。方法:选择47例患有乙型肝炎自然流产史的孕妇,采用荧光定量PCR法检测HBV-DNA含量,分为103~5(Ⅰ组)和>105(Ⅱ组)两组;选取15例健康体检者作为对照组。对上述各组人员进行外周血淋巴细胞染色体制作,G显带核型分析和同步银染法观察染色体着丝粒点变异情况,分析变异率与HBV-DNA含量的关系。结果:前两组外周血淋巴细胞染色体着丝粒点变异率均高于正常对照组,Ⅰ组与对照组、Ⅰ组与Ⅱ组之间均有显著性差异,Ⅱ组与对照组有极显著性差异,病毒含量越高,变异率也越大。结论:乙型肝炎病毒可引起外周血染色体着丝粒点变异,变异率随病毒含量的升高而增加,从而导致染色体不分离,提示乙肝患者Cd的变化是导致流产的重要原因之一。
Objective:To explore the causes of spontaneous abortion by detection of HBV-DNA content in peripheral blood of HBV infected pregnant women with spontaneous abortion and analyzing variation of centromere of chromosome in lymphocytes.Methods:47 HBV infected pregnant women with the history of spontaneous abortion were selected, HBV-DNA content was detected by fluorescence quantitative PCR, then the pregnant women were divided into 103~5group (Ⅰ group) and105 group (Ⅱ group); 15 healthy women were selected as control group; chromosomes in lymphocytes of peripheral blood were prepared, G binding karyotype analysis and synchronous silver staining were used to observe the variation of centromere, the relationship between rate of variation and HBV-DNA content was analyzed.Results:The variation rates of centromere of chromosome in lymphocytes of peripheral blood in the former two groups were significantly higher than that in control group; there was significant difference among I group, II group and control group, the rate of variation increased with the increase of HBV-DNA content.Conclusion:HBV can induce the variation of centromere of chromosome in peripheral blood, the rate of variation increases with the increase of HBV-DNA content, which induces chromosome segregation and indicates that change of centromeric dot is an important cause of abortion.
