摘要
目的:为研究HSV-tk/GCV系统对涎腺腺样囊性癌细胞株(ACC-M)细胞的体外杀伤作用。方法:采用MTT法和细胞计数法对GCV在不同剂量和不同作用时间时细胞的生存率进行研究。结果:HSV-tk/GCV系统能有效地杀伤ACC-M细胞,杀伤呈时间依赖性和剂量依赖性,这种作用有比较明显的旁杀伤效应,旁杀伤效应的强弱与细胞间的接触程度呈正比关系。结论:HSV-tk/GCV系统能有效地杀伤ACC细胞,可望用于ACC的治疗。
Objective: To study the effects of HSV tk/GCV system on adenoid cystic carcinoma(ACC) cells in vitro. Methods: Adenoid cystic carcinoma cells transduced HSV tk gene were treated with ganciclovir(GCV) in different concentrations and at different times, the cell killing was detected with MTT assay. Results: HSV tk/GCV system can kill the ACC M cells greatly in dose dependent and time dependent way, there are apparent bystander effect among cell killing. Conclusions: The HSV tk/GCV system can be used to treat ACC.
