摘要
目的探讨放射免疫介导再障小鼠骨髓中转录因子T-bet的表达及其意义。方法建立再障小鼠模型,取再障和正常组小鼠骨髓单个核细胞(BMMNC),在终浓度为15拉g/mL植物血凝素(PHA)条件下培养,用双抗体夹心ELISA方法检测小鼠骨髓淋巴细胞培养上清Thl及Th2类细胞因子IL-2、IFN-γ和IL.4、IL.10的浓度水平;用RT-PCR技术检测小鼠骨髓转录因子T-betmRNA表达强度。结果再障小鼠骨髓BMMNC培养上清中细胞因子IL-2、IFN-γ水平均显著高于正常组(P〈0.01),而IL.4、IL-10水平均明显低于正常组(P〈0.01);转录因子T-betmRNA表达也明冠高于正常组(P〈0.01)。结论T-bet表达增强可能是再障小鼠出现Th1细胞优势分化状态的重要原因之一。
Objective To investigate the transcription factor T-bet in bone marrow in immune mediated aplastic anemia(AA) mice. Methods Established model of AA mice. Mononuclear cells were isolated from bone marrow incubated with phytohemagglutinin (PHA)(1 5 μg/mL) in vitro.IL-2 and IFN- γ in cultured supernatant were detected by EL1SA.The expression level of transcription factor T-bet was assayed by RT-PCR. Results In AA mice,the level of IL-2 and IFN- γ were much higher than normal (P〈0.01 ),but IL-4,IL- 10 were lower than normal(P〈0.0 1).And the expression of T-bet mRNA was also significantly higher than normal. Conclusion Shifted development of Th toward to Thl in AA mice,and T-bet significantly over expression may be the most important reason.
