摘要
目的探讨外周血单个核细胞(PBMCs)端粒酶在急性川崎病(KD)患儿中表达的活性及其临床意义。方法选择本院2005年3月-2008年12月收治的2个月~6岁的急性KD患儿64例[KD组,其中有冠状动脉损害(CAL)亚组25例,无CAL亚组39例]及同期在本院体检的5个月~7岁健康儿童52例(健康对照组),采用Roche端粒酶PCRELISA法检测其PBMCs,同时检测KD患儿外周血WBC计数、ESR及血清CRP水平。应用SPSS11.0软件对2组结果进行统计学分析。结果 KD组患儿PBMCs端粒酶表达阳性率为32.8%(21/64例),健康对照组为15.4%(8/52例),2组比较差异有统计学意义(χ2=4.65,P<0.05)。KD患儿有CAL亚组PBMCs端粒酶表达阳性率(40.0%,10/25例)高于无CAL亚组(28.2%,11/39例),但二者比较差异无统计学意义(χ2=0.96,P>0.05)。急性期KD患儿PBMCs端粒酶活性阳性组与阴性组炎性指标比较,其差异无统计学意义。结论急性KD患儿外周血淋巴细胞端粒酶表达活性增高可能与KD的发生及发展有关。
Objective To study the telomerase expression in peripheral blood mononuelear cells(PBMCs) in children with acute Kawasaki disease (KD) and its clinical significance. Methods The PBMCs of 64 children with acute KD [ 25 cases of them with coronary artery lesions ( CAL), while the rest without ] from 2 months to 6 ),ears old admitted into Jiangxi Children's Hospital from Mar. 2005 to Dec. 2008 and those of 52 sex -age -matched healthy children (healthy control group) from 5 months to 7 years old were all assayed by Roche telomerase polymerase chain reaction enzymelinked immunosorbent assay( PCR ELISA). WBC, ESR and CRP were also detected. SPSS 11.0 software was used to analyze the data. Results The telomerase expression fi'equency of PBMCs in children with KD was 32.8% (21/64 cases) ,while that in healthy control group was only 15.4% (8/52 cases) ,the difference between the 2 groups was significant (χ^2 = 4.65, P 〈0.05). The frequency of the expression in KD children with CAL was higher than that in non - CAL KD cases,which was 40.0% ( 10/25 cases) and 28.2% (11/39 cases) respectively,but there was no significant difference (χ^2 = 0.96,P 〉 0.05). There were no significant difference of WBC, ESR and CRP between the telomerase of PBMCs positive group and negative group. Conclusions The higher frequency of telomerase expression in peripheral blood lymphoeytes might be related to the development and progression of KD.
