摘要
PTEN是迄今为止人类发现的第一个具有磷酸化酶功能的抑癌基因,PTEN功能的缺失与肿瘤的发生密切相关,该基因异常将导致蛋白表达下降从而失去抑癌作用。目前,有关PTEN基因的研究已经涉及到了其治疗研究,主要是通过转基因技术将野生型PTEN基因导入恶性肿瘤基因组中,替换肿瘤细胞中突变的PTEN基因,发挥正常PTEN基因的抑癌功能。文章就当前PTEN的发现、结构、作用机制与肿瘤的关系及其基因治疗的研究进行综述。
PTEN is the first tumor suppressor gene to be found which has the phosphorylase function so far. PTEN functional loss is closely related with the tumor. This genetic disorder will cause the protein expression decreased, then will lose tumor suppressor function. Currently, the PTEN gene's research has involved in its treatment research. The wild PTEN gene in the malignant tumor genome team replaces the mutational WEN gene in cancer cells mainly by the transgene technology and displays normal PTEN' s tumor suppressor function. Now we makes a summary on the current PTEN discovery, structure, function mechanism and the relationship between the tumor and its gene therapy.
