摘要 目的观察重症急性胰腺炎(severe acute pancreatitis,SAP)大鼠胰腺组织Gi_2和Gq蛋白表达量的改变,探讨在SAP发病过程中血小板活化因子(platelet activating factor,PAF)的细胞信号转导机制。方法 Wistar雄性大鼠60只,随机分为阴性对照组和SAP模型组(SAP组)。以5%牛磺胆酸钠逆行注入主胰管制成SAP模型,阴性对照组开腹后仅翻动十二指肠并触摸胰腺数次关腹。制模后于相应时相点处死动物,取血取组织,ELISA方法检测血浆PAF含量,Western印迹法检测胰腺组织Gi_2/Gq蛋白质的动态变化。结果 SAP组Ci_2和Gq蛋白表达量均较阴性对照组显著增加(P<0.05)。结论 PAF可能通过Gi_2和Gq蛋白,经cAMP-PKA和IP_3-Ca^(2+)途径完成细胞信号转导,导致炎性反应延续并放大。 Objective To study expressions of Gi2 protein and Gi2 protein in pancreas of SAP rats and discover the intracellular signal transduction mechanism of PAF in SAP. Methods Wistar rats were randomly divided into two groups: model group(SAP) and negative control group(NC). SAP was induced by retrograde infusion of 5% sodium taurocholate into the pancreatic duct in Wistar rats. For NC group, the duodenum was moved and the pancreas was palpated the seeral times during the laparotomy , After operation, animals were sacrifieed for detection of plasma cytokines by ELISA and the values of Gi: and Gq proteins in pancreatie tissue were de- termined by Western blotting. Results The values of Gi2 and Gq proteins in SAP group increased significantly compared with NC group (P 〈 0.05). Conelusions There is probably a path to intracellular signal transduction by cAMP - PKA and IP3 - Ca2 + through Gi: and Gq proteins,leading to prolongation and aggravation of inflammatiory response.
