蛛网膜下腔注射pEGFP-C3-hPPE的表达部位及生物学作用

摘要 目的观察蛛网膜下腔注射pEGFP-C3-hPPE后脑啡肽表达的部位及生物学作用。方法取SD大鼠30只随机分为实验组、空质粒对照组与对照组,实验组注射pEGFP-C3-hPPE,空质粒组注射pEGFP-C3,对照组仅作假手术,注射后2 d开始观察各组大鼠的痛觉热敏变化,8 d后注射纳洛酮观察对脑啡肽的拮抗作用,并通过绿色荧光蛋白的表达观察质粒被组织细胞摄取并表达的部位。结果实验组大鼠自术后4 d起痛觉热敏反应时间明显延长,大鼠软脑膜、软脊膜及脊神经外膜上发现绿色荧光蛋白的表达。结论 pEGFP-C3-hPPE能被大鼠软脑膜、软脊膜上皮细胞摄取并表达镇痛的生物学作用。 Objective To observe enkephalin's expressive locations and biologic effects after injecting plasmid pETFP-C3-hPPE into rat's subarachnoid space. Methods 30 rats were divided into experiment group,blank plasmid control group and control group,injected plasmid pETFP-C3-hPPE,plasmid pETFP-C3 and sham operation respectively.After 2 days,observe every rat's algaesthesis temperature sensitive changes.After 8 days,injecting allylnoroxymorphone into rats as well as observing its antagonism against enkephalin and detect green fluorescent protein to determine plasmid's rhagiocrine cell uptook and expressed locations. Results After operation 4 days,experimental group rats prolong their algaesthesis temperature sensitive reaction time obviously and green fluorescent protein are found in cerebral pia mater,spinal pia mater and spinal nerves respectively. Conclusion Plasmid pETFP-C3-hPPE can be uptook by rat's epithelial cell of pia mater and revealed paregoric biologic effects.
