摘要
采用RT-PCR的方法,对水稻条纹叶枯病毒天津分离物的病害特异性蛋白编码基因进行了扩增,将其克隆到pUCm-T载体后进行序列测定与分析.结果表明,该病毒分离物的病害特异性蛋白编码基因的开放阅读框由537个碱基组成,编码的蛋白含178个氨基酸.与GenBank中已有的病害特异性蛋白编码基因进行序列比对,其与分离自北京双桥的SQ、河南原阳的YY、辽宁盘锦的PJ翻译产物只有143位的一个氨基酸的差异.
Full-length cDNA of gene encoding disease-specific protein (SP) of Rice stripe virus(RSV) isolated from Tianjin(TJ) was amplified by RT-PCR. The cDNA was cloned into pUCm-T vector and sequenced. Sequence analysis showed that open reading frame (ORF) of gene encoding SP of this isolate contains 537 nt encoding 178 amino acids. Compared with sequences of genes encoding SP in GenBank, only one amino acid in position 143 is different from SQ isolated from Shuangqiao, Beijing, YY isolated from Yuanyang, Henan and PJ isolated from Panjing, Liaoning.
