摘要
目的:研究纳米雄黄对肺癌A549细胞的凋亡诱导作用。方法:采用机械研磨法制备纳米雄黄。以肺癌A549细胞株为靶细胞,采用MTT法检测细胞的增殖活性,AnnexinV/PI双染色法检测A549细胞的凋亡,流式细胞术检测BAX蛋白和Bcl-2蛋白表达、Caspase-3活性、P53蛋白的表达。结果:纳米雄黄可显著抑制A549细胞的增殖,50μg/mL和100μg/mL的纳米雄黄处理48h后,A549细胞凋亡率显著增高为12.53%和69.19%;20μg/mL和50μg/mL的纳米雄黄作用24h,A549细胞中活性caspase-3由对照的(2.25±0.17)%增高到(3.84±0.63)%和(7.35±0.33)%;P53蛋白表达总体增高但加药48h高剂量组也有所降低;Bax蛋白表达由对照的(79.50±0.50)%增高到(92.45±0.35)%和(96.9±0.00)%;Bcl-2蛋白表达由对照的(77.85±2.15)%增高到(87.10±10.5)%和(83.1±4.6)%。结论:纳米雄黄可有效诱导肺癌A549细胞发生凋亡。
Objective:To explore apoptosis-inducing effects of realgar nanoparticle(nano-realgar) on lung cancer A549 cells.Methods: Preparation of nano-realgar was mechanical milled using a high-energy planetary ball mill.MTT colormetric assay was used to detect the proliferating activity of A549 cells,and the apoptosis of the A549 cells was assed with Annexin V/PI double staining.Flow cytometry(FCM) was employed to assess the expression of intracellular BAX,bcl-2 and P53,and the activity of Caspase-3.Results: Nano-realgar significantly inhibited the proliferation of A549 cells,Treated with 50 g/mL and 100 g/mL of nano-realgar for 48h,the apoptosis rate of the A549 cells by Annexin V/PI staining was increased to 12.59% and 69.19%,respectively,the activity of Caspase-3 significantly was enhanced,from(2.25±0.17)% to(3.84±0.63)% and(7.35±0.325) %.P53 protein expression increased,but the 48h and high dose group decreased.The Bax protein expression in target cells was increased from(79.50±0.50)% to(92.45±0.35)% and(96.90±0.00)%.Bcl-2 protein expression in target cells was increased from(77.85±2.15)% to(87.10±10.5)% and(83.1±4.6)%.Conclusion: Realgar nanoparticle significantly induces the apoptosis of lung cancer A549 cells.
