摘要
以I-101×84K杨为材料,对其组培快繁进行了研究。试验结果表明,MS为合适的基本培养基,根据正交试验结果,诱导I-101×84K杨腋芽增殖的最佳培养基MS+6-BA0.3 mg.L-1+NAA0.3 mg.L-1+蔗糖30 g.L-1+琼脂6 g.L-1;生根培养基以1/2MS+NAA0.02 mg.L-1+蔗糖15 g.L-1+琼脂6 g.L-1为宜,生根试管苗炼苗、移栽后成活率可达90%。对试验过程中的玻璃化问题进行了初步的研究,发现大量元素浓度过高和多次继代可引起试管苗的玻璃化。
Tissue culture for rapid propagation was conducted by using I-101×poplar 84K(Populus alba×(P.alba×P.glandulosa)) stems as the explants.The results showed that the optimal basic medium was the MS.Based on orthogonal experiment,the optimal proliferation medium for the axillary buds was MS+0.3 mg·L^-16-BA +0.3 mg·L^-1 NAA +cane sugar 30 g·L^-1+ agar 0.6 g·L^-1.For rooting,the optimal medium was 1/2MS + 0.02 mg·L^-1 NAA + cane sugar 15 g·L^-1 + agar 0.6 g·L^-1.90% of survival rate of the cultured seedlings were achieved after hardening and transplanting processes.It was found that higher concentrations of macroelements and repeated subculture may cause plantlet vitrification.
