摘要
目的探讨凝血酶预处理(TPC)对脑出血(ICH)大鼠神经细胞凋亡及半胱氨酸蛋白酶-3(caspase-3)表达的影响。方法 60只成年雄性SD大鼠随机平均分为10组:假手术(SO)组、TPC 3 h组、TPC6 h组、TPC 12 h组、TPC 1 d组、TPC 3 d组、TPC 7 d组、TPC 14 d组、TPC 21 d组及ICH组。TPC各组大鼠与ICH组大鼠予以右侧尾状核注射凝血酶10 U诱发脑出血,SO组大鼠予以同部位注射生理盐水50μl,用dUTP缺口末端标记法(TUNEL)染色及免疫组化染色法观察神经细胞凋亡及caspase-3表达的情况。结果 ICH组与TPC各组凋亡细胞及caspase-3表达均明显多于SO组(均P<0.01),TPC 12 h、TPC 1 d、TPC 3 d、TPC 7 d、TPC 14 d和TPC 21 d组细胞凋亡及caspase-3阳性表达明显少于ICH组(均P<0.05),其中TPC 7 d组凋亡细胞数及caspase-3阳性表达最少(P<0.05~0.01)。结论 TPC可减少ICH所致的神经细胞凋亡及caspase-3表达,其中以TPC 7 d的作用最强。
Objective To study the effect of thrombin preconditioning(TPC) on neurocytes apoptosis and expression of caspase-3 in intracerebral hemorrhage(ICH) rats.Methods 60 adult male SD rats were randomly divided into 10 groups: sham operation(SO) group,TPC 3 h group,TPC 6 h group,TPC 12 h group,TPC 1d group,TPC 3 d group,TPC 7 d group,TPC 14 d group,TPC 21 d group and ICH group.All rats from TPC groups and ICH group received 10 U thrombin injection in the right caudate nucleus to induce ICH.SO group rats received 50 μl saline in the same site.TUNEL and immumohistochemical methods were used to observe the neurocytes apoptosis and expression of caspase-3.Results Compared with SO group,neurocytes apoptosis and expression of caspase-3 in all TPC groups and ICH group increased significantly(all P〈0.01).Compared with ICH group,neurocytes apoptosis and expression of caspase-3 in TPC 12 h,TPC 1 d,TPC 3 d,TPC 7 d,TPC 14 d and TPC 21 d groups reduced obviously(all P〈0.05),and TPC 7 d group showed the least(P〈0.05~0.01).Conclusions TPC can decrease the neurocytes apoptosis and the expression of caspase-3 which caused by ICH,and the protective effect in TPC 7 d is the best.
