摘要
目的比较日本血吸虫非适宜宿主东方田鼠和SD大鼠特异性DNA序列的差异。方法提取东方田鼠、SD大鼠、C57BL/6小鼠和KM鼠肝脏DNA,根据东方田鼠特异性DNA序列片段(GenBank登录号:AF277394)设计引物,PCR方法扩增以上4种鼠的基因组特异性DNA,琼脂糖凝胶电泳鉴定特异条带,纯化后的PCR产物克隆到pGEM-T Easy载体,进行DNA序列分析。结果 PCR扩增东方田鼠、SD大鼠DNA分别得到520bp左右的特异扩增片段,同源性为99%。C57BL/6小鼠和KM鼠没有扩增出此特异片段。结论日本血吸虫非适宜宿主东方田鼠和SD大鼠之间特异性DNA序列有高度的同源性。
Objective To compare the difference between the DNA sequences of Microtus fortis and SD rats,non-permissive hosts of Schistosoma japonicum.Methods The DNAs were extracted from the liver tissues of M.fortis,SD rats,C57BL/6 mice and KM mice,respectively.The primers were designed according to the specific DNA sequences of M.fortis（GenBank accession number：AF277394）,and PCR was carried out for amplifying the genome DNAs of M.fortis,SD rats,C57BL/6 mice and KM mice.Following the identification of agarose gel electrophoresis,the purified DNA fragments were cloned into the pGEM-T Easy vector for sequenceing.Results Two specific DNA fragments of around 520 bp were obtained for M.fortis and SD rats and they showed a high homology of 99% with each other.However,no fragments were observed of the PCR products of C57BL/6 and KM mice.Conclusion The specific DNA fragments of the two non-permissive hosts of S.japonicum（M.fortis and SD rats）show a high homology.
