摘要
目的用乙烯硫脲诱发妊娠大白鼠产生泄殖腔畸形动物模型,观察其肛周肌肉病理改变。方法取泄殖腔畸形鼠仔,正中矢状面连续切片,用LuzexF图像分析仪测内、外括约肌及耻骨直肠肌面积。用肌球蛋白ATP酶染色,观察鼠仔盆底横纹肌复合体中Ⅰ、Ⅱ型肌纤维发育情况。结果各种泄殖腔畸形内括约肌面积较正常鼠增大,后壁更明显(P<0.05);肛门外括约肌面积较正常鼠明显小;低位泄殖腔畸形耻骨直肠肌面积较正常鼠增大,而中高位其面积较正常鼠小。泄殖腔畸形鼠仔盆底横纹肌复合体中Ⅰ型肌纤维占比例明显减少(P<0.05)。结论泄殖腔畸形动物模型病理改变与人类极相似。
Objective To study the pathological changes of peri anal musculature in Wistar rats with ethylenethiourea induced cloacal anomalies.Methods Serial sagittal sections were performed on fetuses which developed cloacal anomalies after being exposed to ethylenethiourea in utero. Puborectalis, internal and external anal sphincters were measured with Luzes Fimage analyzer. The striated muscles of pelvic floor were stained for type Ⅰ and type Ⅱ fibers.Results Of the cloacal anomalies group, the internal anal sphincter was significantly thicker than that of the normal, especially over the posterior aspect ( P < 0.05 ). The size of puborectalis muscle was larger than that of normal in low type anomalies, but smaller than normal in intermediate and high types of anomalies. The proportion of type Ⅰ fibers in striated muscle complex was significantly lower ( P < 0.05 ).Conclusions The pathological changes in this animal model are similar to those seen in human beings. Hence the model provides a useful tool in research of cloacal anomalies.
