摘要
被孢霉是花生四烯酸发酵生产的最佳菌种,但在长期的生产过程中,被孢霉菌种极易发生退化,从而导致许多高效优良的性能丧失。以1株高山被孢霉ME-1为研究对象,在PDA培养基的基础上,添加不同的微量营养因子(蛋白胨、酵母膏、牛肉膏)作为复壮培养基,考察不同的复壮载体对菌株复壮效果的影响。实验结果表明,最佳复壮培养基为PDA+BE+PE培养基,效果十分明显。复壮菌株发酵性能稳定,其生物量、总油脂产量和ARA产量分别比原始菌株提高了25%、25%和30%。
Mortierella was the preferred strain for arachidonic acid(ARA) fermentation,but Mortierella species was very prone to degradation in the long-term ARA production.Mortierella alpina ME-1 was seclected as our research target,and different micro-nutritional factors(peptone,yeast extract,beef extract) was added to basical PDA medium as the rejuvenation medium.The purpose of this paper was to investigate the effects of different rejuvenation medium for the separation of high-yield ARA strain.The experimental results showed that the best medium rejuvenation for high-yield ARA strain separation was the PDA +BE+PE medium,and the effect was quite obvious.The strain obtained from was extremely stable in ARA fermentation.Compared with the original strain,the value of biomass,oil concentration and ARA concentration increased by 25%,25% and 30%,respectively.
