摘要
目的建立大鼠小脑外形三维重构的可视化数据集。方法采用小脑连续切片,同时建立采集切片后的图像数据,切片经尼氏染色后,用数码相机采集图像,尼氏染色后的图像与切片时采集的图像配准,进行图层合并,获取原始图像数据库,并对数据库中的数据进行人工配准及分割,应用Amira4.1.1软件对分割后的数据库进行三维重建。结果获得大鼠小脑外形微细结构信息的数据库。结论本法是将传统的由组织切片进行三维重建技术与用于虚拟人研究的机床铣切及同步数据采集系统相结合的新方法 ,适用于组织微细结构的三维重建。
Objective To establish a detailed digitalized dataset for the rat cerebellum by 3D reconstruction. Methods The rat cerebellum was sectioned serially and stained with Nissel's method,then photographed with camera.The original image dataset was collected,aligned and segmented manually.The segmented structures were reconstructed using Amira 4.1.1. Results The original dataset of the detailed morphological structure of rat cerebellum was achieved. Conclusion 3D digital model of cerebellum is successfully obtained by combining digital human techniques and histochemical stain.This method is useful for 3D reconstruction of tissue microstructure.
