摘要 目的从细胞凋亡的角度探讨当归多糖(APS)抑制HaCaT细胞增殖的机制。方法用细胞体外培养技术、台盼蓝拒染法与流式细胞术检测APS对HaCaT细胞增殖的影响,电镜观察细胞形态变化。结果细胞生长曲线显示25~2500mg/L的APS对HaCaT细胞有显著抑制作用,且呈剂量效应关系。流式细胞术结果显示250mg/L作用后S/G2+M期HaCaT细胞数减少,G0/G1期细胞增多。结论 APS对HaCaT细胞的增殖具有显著抑制作用,APS通过阻止HaCaT细胞进入S期而抑制其DNA合成。 Objective To explore the mechanism of angelica polysaccharide (APS)inhibiting proliferation of HaCaT cells from the angulation of apoptosis. Methods Techniques of cell culture were used in this experiment. The effect of APS on proliferation of HaCaT cells was examined by trypan blue staining and flow cytometry. The change of cell form was observed by scanning electron microscope. Results Cell growth curve showed 25-2500mg/LAPS had significant inhibition action on HaCaT cells with dose-dependent manner. Flow cytometry result showed the S phase and G2+M phase HaCaT cells decreased and the G0/G1 phase HaCaT cells increased after 250mg/L APS had been given. Conclusion APS could significantly inhibit proliferation of HaCaT cells. APS inhibits its DNA synthesis through preventing HaCaT cells to enter S phase.
